I use both, but Twitter much more often than Plurk - and I use them in slightly different ways that are hard to define. There is some overlap in who uses which one, but I get the sense that it's a little younger and lighter on Plurk, more business use and serious networking at Twitter. But of course, all that depends on who you hang out with (follow and/or friend) on each service. If you plan on dipping in a toe - best way is probably to pick the one where you know the most people who might have something to say that you might want to hear!
